So I’m sick to death of my deadlocks.
The sliding door and barn mechanisms have been deactivated and I’m not sure what is missing. The handles don’t work so I can only open with the one key that came with the van. If I lose it, I’m screwed!
I’d rather just have the doors on the central locking. Nothing gets left in it, so not worried about break ins.
Can I simply replace the units? Or is there coding involved?

First I would get a spare key, just in case and you never know when you will need it.
One or both handles have been taken out and placed back that you know or it just happens from the one to the other moment? If it’s the first, start to check if everything is placed correct back. After wrapping a caddy I know that I have to try to get it working in fact of just place them back otherwise it can result in the same as you describe.
